چکیده

The importance of two of the thiol groups of yeast triosephosphate dehydrogenase for its activity haa been shown by the abolition of activity and diphosphopyridine nucleotide binding by the addition of 2 equivalents of p-chloromercuribenzoate (PCMB) (1, 2). The reaction of methyl mercury nitrate with this enzyme has been found to be anomalous in the sense that this mercury compound, unlike PCMB, does not react first with the enzyme sulfhydryl groups which are required for activity. These results and some determinations of the total numbers of sulfhydryl groups available in the enzyme after various treatments are presented here.

Transfer reactions of acetyl phosphate catalyzed by glyceraldehyde-3-phosphate dehydrogenase.

Glyceraldehyde-3-phosphate dehydrogenase catalyzes the oxidative phosphorylation of acetaldehyde (1) and a number of other aliphatic aldehydes (2, 3) and also has a transacetylating activity (4). In the present paper the reactions of acetyl phosphate which characterize the transacetylating activity of the enzyme are presented. These reactions include (a) the phosphate exchange between acetyl ph...

On the occurrence, equilibria, and site of acyl-enzyme formation of glyceraldehyde-3-phosphate dehydrogenase.

Researches in several laboratories have given evidence that catalysis of aldehyde oxidation by glyceraldehyde-3-phosphate (G-3-P) dehydrogenase involves formation of an acyl-enzyme intermediate which is subsequently cleaved by inorganic orthophosphate (Pi) to give an acyl phosphate (l-5). Recently Warburg et al. (6) have expressed doubt that present evidence warrants acceptance of the acyl-enzy...

A solid support for affinity chromatography that covalently binds thiol groups via a cleavable connector arm.

Preparation of an agarose derivative (MPE-agarose) containing a maleimido group which is attached to agarose via a cleavable phenyl ester linkage is described. MPE-agarose was shown to react with the thiol groups in glutathione, bovine serum albumin, bovine hemoglobin, and yeast and rabbit muscle glyceraldehyde 3-phosphate dehydrogenase. Treatment of the resulting agarose-linked compounds for 1...

Alkylation of glyceraldehyde-3-phosphate dehydrogenase with haloacetylphosphonates. An unusual pH-dependence.

Two new alkylating reagents, chloro- and bromo-acetylphosphonate, were found to be very effective thiol-blocking reagents. The pH-dependence of the reaction of BAP with 2,4-dinitrothiophenol (25 degrees C, I 0.5) shows a tailing bell-shaped curve (with a plateau at high pH) characteristic of two ionizing groups: the thiol group (pKa 3.2) and the phosphonate group (pKa2 4.6). The rate constant f...

A novel diazonium-sulfhydryl reaction in the inactivation of yeast alcohol dehydrogenase by diazotized 3-aminopyridine adenine dinucleotide.

Diazotized 3-aminopyridine adenine dinucleotide has been found to modify four sulfhydryl groups per molecule of enzyme during the complete inactivation of yeast alcohol dehydrogenase. The reaction of sulfhydryl groups was indicated by titration studies with 5,5-dithiobis(2-nitrobenzoic acid) as well as isolation and quantitation of the cysteinyl derivative released by acid hydrolysis of the mod...
